ABSTRACT

The present study aimed to investigate the composition of the terpene synthase(TPS) gene family in Gynostemma pentaphyllum and its role in abiotic stresses. The G. pentaphyllum TPS gene family was identified and analyzed at the genome-wide level using bioinformatics analysis, and the expression patterns of these family members were analyzed in different tissues of G. pentaphyllum as well as under various abiotic stresses. The results showed that there were 24 TPS gene family members in G. pentaphyllum with protein lengths ranging from 294 to 842 aa. All of them were localized in the cytoplasm or chloroplasts and unevenly distributed on the 11 chromosomes of G. pentaphyllum. The results of the phylogenetic tree showed that the G. pentaphyllum TPS gene family members could be divided into five subfamilies. As revealed by the analysis of promoter cis-acting elements, TPS gene family members in G. pentaphyllum were predicted to respond to a variety of abiotic stresses such as salt, low temperature, and dark stress. The analysis of gene expression patterns in different tissues of G. pentaphyllum revealed that nine TPS genes were tissue-specific in expression. The qPCR results showed that GpTPS16, GpTPS17, and GpTPS21 responded to a variety of abiotic stresses. This study is expected to provide references in guiding the further exploration of the biological functions of G. pentaphyllum TPS genes under abiotic stresses.

ABSTRACT

Abstract Purpose: To investigate the role of PI3k/Akt signal pathway in the protective effects of propofol on intestinal and lung injury induced by intestinal ischemia/reperfusion(I/R). Methods: Male Sprague-Dawley rats were subjected to 45 min of ischemia by occluding the superior mesenteric artery and to 2h of reperfusion to establish the model of I/R. Twenty four rats were randomly divided into four groups: Sham, intestinal I/R (II/R), propofol (P), wortmannin (W). In groups P, W, propofol was injected intravenously and continuously at the onset of reperfusion via infusion pump. PI3K inhibitor (wortmannin) was administered intravenously in group W 25 min before ischemia. Intestinal tissues and lung tissues were obtained for determination of histologic injury, wet/dry weight ratio, malondialdehyde (MDA) levels, superoxide dismutase (SOD) and myeloperoxidase (MPO) activities. Meanwhile, the expressions of caspase-3 and phosphorylated Akt (p-Akt) in intestines and lungs were detected by western blot. Results: Propofol treatment alleviated intestinal and lung morphological changes which were observed in II/R group,Moreover, wet/dry weight ratio, the MDA level, MPO activity and expression of caspase-3 were significantly decreased whereas the SOD activity and p-Akt expression were significantly increased. Notably, the protections were significantly reversed by pretreatment of wortmannin. Conclusion: PI3K/Akt pathway activation play a critical role in the protective effects of propofol on intestinal and lung injury induced by ischemia/reperfusion.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the effects of salvianolate on myocardial infarction in a murine in vivo model of ischemia and reperfusion (I/R) injury.</p><p><b>METHODS</b>Myocardial I/R injury model was constructed in mice by 30 min of coronary occlusion followed by 24 h of reperfusion and pretreated with salvianolate 30 min before I/R (SAL group). The SAL group was compared with SHAM (no I/R and no salvianolate), I/R (no salvianolate), and ischemia preconditioning (IPC) groups. Furthermore, an ERK1/2 inhibitor PD98059 (1 mg/kg), and a phosphatidylinositol-3-kinase (PI3-K) inhibitor, LY294002 (7.5 mg/kg), were administered intraperitoneal injection (i.p) for 30 min prior to salvianolate, followed by I/R surgery in LY and PD groups. By using a double staining method, the ratio of the infarct size (IS) to left ventricle (LV) and of risk region (RR) to LV were compared among the groups. Correlations between IS and RR were analyzed. Western-blot was used to detect the extracellular signal-regulated kinase 1/2 (ERK1/2) and protein kinase B (AKT) phosphorylation changes.</p><p><b>RESULTS</b>There were no significant differences between RR to LV ratio among the SHAM, I/R, IPC and SAL groups (P>0.05). The SAL and IPC groups had IS of 26.1%±1.4% and 22.3%±2.9% of RR, respectively, both of which were significantly smaller than the I/R group (38.5%±2.9% of RR, P<0.05, P<0.01, respectively). Moreover, the phosphorylation of ERK1/2 was increased in SAL group (P<0.05), while AKT had no significant change. LY294002 further reduced IS, whereas the protective role of salvianolate could be attenuated by PD98059, which increased the IS. Additionally, the IS was not linearly related to the RR (r=0.23, 0.45, 0.62, 0.17, and 0.52 in the SHAM, I/R, SAL, LY and PD groups, respectively).</p><p><b>CONCLUSION</b>Salvianolate could reduce myocardial I/R injury in mice in vivo, which involves an ERK1/2 pathway, but not a PI3-K signaling pathway.</p>

ABSTRACT

Objective To study the association between serum concentrations of total testosterone levels and coronary heart disease (CHD) in the postmenopausal women. Methods The study was designed as a case-control study.394 postmenopausal female patients were selected from Cardiology Center of the First Affiliated Hospital of Xinjiang Medical University. The case group included 183 women patients with CHD aged (62.7±8.0) years, the control group,211 women with normal coronary aged (60.0 ± 8.8) years. Blood samples were collected to determine total testosterone, fasting glucose and lipid profile. CHD severity was expressed as the numbers of coronary arteries that had a stenosis ≥50%.According to the level of testosteron, all cases were divided into 4 groups by interquartile range method:Q1<3.5 nmol/L(n=190),3.5 nmol/L≤Q2 <10.4 nmol/L(n=64),10.4 nmol/L≤Q3 <26.0 nmol/L(n=120) and Q4≥26.0 nmol/L(n=20).The association between the serum total testosterone levels and severity of coronary atherosclerosis was analyzed. Results The average total testosterone was higher in case group than in control group[(10.4 ± 24.3 ) nmol/L vs. ( 6.9 ± 17.4 ) nmol/L,Z =0.79,P =0.555].In Quantitative adjusted models, higher levels of total testosterone had strong correlation with CHD,Q4 incidence of CHD (75.0%,15 cases) was significantly higher than Q1 (46.8%,89 cases),Q2 (40.6%,26 cases)and Q3 (44.2%,53 cases) (x2 =7.69,P=0.048).After adjustment for other risk factors, women in the top quartile of total testosterone levels had a more than 3-fold increase in odds of CHD(OR=3.47,95%CI:1.06-11.32,P<0.05).In addition, the serum concentrations of total testosterone level were significantly associated with the severity of CHD (F=12.94,P<0.05). Conclusions Higher levels of total testosterone may be associated with high prevalence and severity of CHD as an independent factor in postmenopausal women.

ABSTRACT

In recent years, there are tremendous economic and social losses across the world because of virus-related diseases. It is well known that Madin-Darby canine kidney (MDCK) cells are easily handled, quickly amplified and efficiently infected with influenza virus. Therefore, they are considered as one of the most important cell lines for the production of influenza vaccine. In this work, we first developed a serum-free adherent culture process for MDCK cells with an in-house prepared serum-free medium MDCK-SFM. Next, we derived a cell line named ssf-MDCK, which was amenable for single-cell suspension culture in the serum-free medium. We found that during serum-free batch culture of MDCK cells, the peak viable cell density and maximum specific growth rate were 3.81 x 10(6) cells/mL and 0.056 h(-1), respectively; 3.6- and 1.6-fold increase compared with those in serum-containing adherent batch culture. In addition, we compared growth and metabolic characteristics of MDCK cells in serum-containing adherent culture, serum-free adherent culture and serum-free single-cell suspension culture. We found that less metabolic by-products were produced in both serum-free cultures. In serum-free single-cell suspension batch culture, the viable cell density was highest. These results are critical for establishing large-scale suspension culture of MDCK cells as subsequent well as large-scale influenza vaccine production.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the association between genetic polymorphism of serum amyloid protein A1 (SAA1) with carotid intima media thickness in a healthy Han Chinese population of Xinjiang.</p><p><b>METHODS</b>A total of 449 healthy Han Chinese participating the cardiovascular risk survey between June 2007 and September 2009 were included, the genotypes of the SAA1 were detected by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). The mean IMT of the right and left common carotid arteries were measured by B-mode ultrasonography.</p><p><b>RESULTS</b>(1) There was strong linkage disequilibrium between rs12218 and rs2229338 (D' = 0.89). (2) The carotid common IMT (CC-IMT) and the carotid bulb IMT (CB-IMT) were similar between the AA genotype (wild genotype) and the GGFAG genotype (mutational genotype) in rs2229338 of SAA1 gene. (3) CC-IMT [(0.081 ± 0.071) cm vs (0.068 ± 0.019) cm, P = 0.01] was significantly thicker in CC + CT genotype (mutational genotype) group than in TT genotype (wild genotype) of rs12218 group and the difference remains significant after adjustment for age, gender, blood pressure, waist circumference, creatinine and high density lipoprotein cholesterol. CB-IMT [(0.085 ± 0.038) cm vs. (0.081 ± 0.052) cm, P = 0.36] was similar between CC + CT genotype and TT genotype of rs12218 groups.</p><p><b>CONCLUSION</b>Our results suggested that the genetic polymorphism of SAA1 might be linked with IMT and rs12218 mutation could serve as a promoting factor for IMT in Han Chinese people.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To compare the clinical efficacy of the infiltration anesthesia with primacaine and the nerve blocking anesthesia with lidocaine for microport extraction of impacted lower third molar. METHODS; 104 chosen patients had both sides of impacted lower third molars extracted in this study. Patients were given local anesthesia with either primacaine or lidocaine randomly at each side, and then underwent microport extraction. Clinical factors including effective proportion (EP), effecting time point (ETP), visual analogue scale of pain (VASp), alteration of systolic pressures (ASP) and analgesia duration (AD) were evaluated statistically by means of paired t-test.</p><p><b>RESULTS</b>The EP of experimental group was higher than the control group (P = 0.024). The ETP of soft tissue and alveoli-dental pulp was (1.04 +/- 0.21), (2.44 +/- 2.60) min in the experimental group, and much earlier than that of the control group (P = 0.002, P = 0.032). The VASp and ASP of experimental group were lower than the control group (P = 0.041, P = 0.018). AD was (103.6 +/- 35.5) min, and higher than the control group (P = 0.04).</p><p><b>CONCLUSION</b>The infiltration anesthesia with primacaine has been proven to be a easier, reliable and quick-acting method. We suggest it an alternative method replacing the 2% lidocaine blocking during microport extraction of impacted lower third molar.</p>

ABSTRACT

Objective To study the correlation between renal function and arterial stiffness of Uighur population over 35 years old in Xinjiang area. Methods From October 2007 to March 2010, four-stage stratified random sampling method was applied to collect a total of 4312 Uighur people over 35 years old in Urumqi, Karamay, Fukang, Turpan area, Hotan prefecture, Yili Kazak autonomous prefecture and Altai prefecture. The data of these 4312 general urban and rural Uygur residents were complete. Simplified MDRD formula was used to calculate the estimate glomerular filtration rate (eGFR). Arm-ankle pulse wave velocity (b-aPWV) of both sides was measured and the average value was used in correlation analysis with eCFR, and then the associated risk factors were adjusted. Results The eGFR was negatively correlated with b-aPWV (r=-0.174, P<0.01). Multiple linear regression showed that after adjusting some risk factors such as age, sex, body mass index, systolic blood pressure, diastolic blood pressure, waist circumference, hip circumference, uric acid, fasting blood glucose, total cholesterol, triglycerides, low density lipoprotein, eGFR was still negatively correlated with b-aPWV (P=0.012). Conclusions In Uygur population over 35 years old in Xinjiang, the glomerular filtration rate is negatively correlated with arterial stiffness.

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical features, treatment methods and outcome of solitary plasmacytoma of cervical spine.</p><p><b>METHODS</b>From January 1995 to December 2007, the data of 23 cases with solitary plasmacytoma of cervical spine was analyzed. There were 16 males and 7 females (mean age 56 years, range: 32 - 76 years). Two cases underwent radiotherapy alone and 21 patients received surgery. According to WBB staging system, surgical procedures were defined as total or subtotal resection (6 cases), appendix resection (4 cases), sagittal resection (3 cases) and total spondylectomy (8 cases). All surgical cases were managed using an anterior approach, posterior approach or combined anterior and posterior approach. The cervical spinal reconstruction was achieved through anterior cervical titanium plate and titanium mesh cage filled with auto iliac graft or bone cement, or anterior and posterior combined instrumented fusion. All patients received radiotherapy as adjunctive therapy.</p><p><b>RESULTS</b>Follow-up of the 23 cases lasted 24.0 - 143.0 months (mean: 64.7 months). Neck pains obviously improved, and nerve compression symptoms disappeared or improved after surgery. Neurological function improved by 1 - 2 grades based on Frankel grading system. All the internal fixations were fused well and stability of the cervical spine was fine and no spine instability could be seen in our series. The bone graft fusion rate was 100%. During the follow-up period, 6 surgical cases had local recurrence and finally progressed to multiple myeloma (MM) and 3 died. Two cases without surgical treatment progressed to MM in 1 year and 1.5 years after confirmed diagnosis. They were given systemic chemotherapy. The other 15 patients had disease-free survival and after surgery and adjunctive radiotherapy. Obvious abnormity were not found in such examinations as M protein, bone marrow aspiration and emission computed tomography or PET-CT examinations.</p><p><b>CONCLUSIONS</b>Solitary plasmacytoma of cervical spine is rarely seen clinically. Surgery is recommended as the primary management for patients with overt bone destruction and spinal instability or neurological dysfunction. Tumor excision with adjunctive radiotherapy can obviously reduce local recurrences and lower the possibility of progression to MM. The patients with progression to MM should receive chemotherapy according to chemotherapy protocol while the prognosis is comparatively worse.</p>

Methods Four-stage selected random samples with maternal age at 35 or over were used to analyze the prevalence and distributing feature of self-reported congestive heart failure in different nationalities, age, sex. The sampled adult population were collected from 6 localities(Urumqi, Kelamayi, Fukang, the Turfan Basin locality, Hetian locality, Yili Hazakh autonomous prefecture), 23 municipalities and 7 locality and 5 autonomous counties in Xinjiang. Results 16 460 people were surveyed. The prevalence rates of overweight and obesity were 36.1% and 26.9% in Han, Uygur and Hazakh population in Xinjiang, respectively from February, 2007. The prevalence rates of overweight and obesity were 41.4% and18.4% in Han population, 34.9% and 28.9% in Uygur population, but 32.8% and 40.1% in Hazakh population. The prevalence rate of overweight and obesity was higher in males(x2= 135.00, P<0.05).The prevalence rates of overweight and obesity were different between different ethnic groups(x2=338.232, P<0.05). The prevalence of overweight was highest in Han population, with the highest seen in Hazakh population. The prevalence rates of overweight and obesity were increasing with age (x2=246.80,P<0.05). The overweight rate in 45-54 year olds and the obesity rate in 55-64 year olds reached their peak values. Results from logistic regression model analyses indicated that the prevalence of overweight and obesity in Xinjiang were statistically associated with age, educational level, jobs, smoking and alcohol consumption. Conclusion The prevalence rates of overweight and obesity were much higher in the population of Xinjiang but different among ethnicities. The prevalence of overweight was the highest in Han male population and the rate of obesity in Hazakh male population was the highest.

ABSTRACT

BACKGROUND:Arterial stiffness is an independent risk factor for cardiovascular disease.Augmentation index(Alx)is an important marker which reflects the systemic arterial stiffness.OBJECTIVE:To investigate Alx and the affecting factors in Xinjiang Uygur health adults.METHODS:A total of 559 healthy adults(251 males and 308 females)with a mean age of(44.95±11.87)years were selected from Hetian,Xinjiang Uygur Autonomous Region. The following indexes of all subjects were measured, including questionnaire, physical examination, blood biochemistry and C-reactive protein(CRP).Central systolic blood pressure(cSBP)and central Alx were measured by Sphygmocor.Pearson correlation and partial correlation analysis were used for two-group analysis of correlation, and multiple regression analysis was used to analyze influential factors for Alx.RESULTS AND CONCULUSION:Alx was greater in female than in male and was positively related to age in both sexes(male:r=0.548,female:r =0.405,P < 0.05),as well as adjusted other influential factors(male:r=0.182,female:r =0.234,P < 0.05).Multiple regression analysis showed that cSBP,systolic blood pressure(SBP),sex, age and CRP were main factors which affected Alx in Uygur healthy adults,cSBP,SBP,age, body mass and CRP were main factors which affected Alx in the male, while cSBP,SBP,age, height and CRP were main factors which affected Alx in the female. Results revealed that Alx was greater in female than in male which was positively related to age in both sexes.Age,cSBP,SBP and CRP were main factors that affected Alx in Xinjiang Uygur healthy adults.

ABSTRACT

Objective To investigate the relationship between stroke and ankle-brachial index (ABI), pulse wave velocity (PWV) in Han, Uighur, and Kazakh populations of Xinjiang. Methods Data was from the investigation of cardiovascular risk survey (CRS) program among different nationalities in Xinjiang from October 2007 to March 2010. A total of 14 618 samples aged over 35 with complete data dimension were surveyed. Results ( 1 ) There were 633 patients with stroke, including 258 Hans (4.48%), 247 Uighurs (5.18%), and 128 Kazakhs (3.13%). The incidence of stroke was significant different in the three ethnic groups (P<0.001). (2)Compared to the Han population, the incidence of stroke was higher ( OR= 1.304) in Uighur but lower (OR=0.794) in Kazakh. (3) PWV was significant different between stroke and non-stroke patients. After adjustment for age, body mass index, systolic blood pressure, triglyceride, total cholesterol and other risk factors, the difference remained significant, indicating that PWV (OR=1.001, P<0.001) might associate with the occurrence of stroke. Conclusion The results of this study showed that the incidence of stroke was significantly different in the three ethnic groups. PWV might associate with the occurrence of stroke.

ABSTRACT

<p><b>AIM</b>To investigate the effect of DAPT (gamma-secretase inhibitor) on the growth of human tongue carcinoma cells and to determine the molecular mechanism to enable the potential application of DAPT to the treatment of tongue carcinoma.</p><p><b>METHODOLOGY</b>Human tongue carcinoma Tca8113 cells were cultured with DAPT. Cell growth was determined using Indigotic Reduction method. The cell cycle and apoptosis were analyzed by flow cytometry. Real-time PCR and Immuno-Fluorescence (IF) were employed to determine the intracellular expression levels.</p><p><b>RESULTS</b>DAPT inhibited the growth of human tongue carcinoma Tca8113 cells by inducing G0-G1 cell cycle arrest and apoptosis. The mRNA levels of Hairy/Enhancer of Split-1 (Hes-1), a target of Notch activation, were reduced by DAPT in a dose-dependent manner. Coincident with this observation, DAPT induced a dose-dependent promotion of constitutive Caspase-3 in Tca8113 cells.</p><p><b>CONCLUSION</b>DAPT may have a therapeutic value for human tongue carcinoma. Moreover, the effects of DAPT in tumor inhibition may arise partly via the modulation of Notch-1 and Caspase-3.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the effects of percutaneous balloon pulmonary valvuloplasty (PBPV) for patients with pulmonary valve stenosis (PS).</p><p><b>METHODS</b>From February 1996 to March 2003, 65 patients with isolated PS were diagnosed by echocardiography and received PBPV in our department, clinical data were analyzed in this study.</p><p><b>RESULTS</b>Age of 65 patients ranged from 1 to 48 years [mean (13.5 +/- 9.3) years]. The pulmonary transvalvular gradient (PTG) was (86.4 +/- 33.6) mm Hg(1 mm Hg = 0.133 kPa) and the right ventricular systolic pressure was (107.5 +/- 36.5) mm Hg before PBPV. Single-balloon valvuloplasty was performed in 41 patients, double-balloon valvuloplasty in 6 patients and Inoue-balloon valvuloplasty in 18 patients. The ratio of balloon/valve ranged from 1.00 to 1.19 in 19 patients, from 1.20 to 1.39 in 42 patients, and greater than 1.40 in 4 patients. The procedure was classified as successful when the RV-PA gradient was < 36 mm Hg post procedure, 6 to 12 months follow up was finished after PBPV by catheterization or echocardiography in 25 patients. Immediate post procedure success rate was 81.5% (53/65). The lowest PTG immediately post procedure was seen in Inoue balloon group and balloon/valve ratio between 1.20 to 1.39 group. Post procedure, tricuspid valve regurgitation was evidenced in 2 patients, reactive right ventricular outflow tract stenosis was shown in 26 patients (13 from Inoue group) and pulmonary regurgitation was detected in 3 patients. The mean PTG was (35.7 +/- 23.9) mm Hg at follow-up. PTG gradually reduced to normal in 6 patients with post procedure PTG > or = 36 mm Hg. Right ventricular outflow tract stenosis was attenuated in all 11 followuped patients. There was no restenosis during follow-up.</p><p><b>CONCLUSION</b>Percutaneous balloon valvuloplasty was effective and safe for treating patients with pulmonary valve stenosis. Superior outcome was linked with balloon/valve ratio between 1.20 to 1.39 and Inoue balloon valvuloplasty in this cohort.</p>

ABSTRACT

Objective The aim of this study was to assess the association between human CYP4F2 gene and myocardial infarction (MI),using a haplotype-based case-control study. A separate analysis on gender was also carried out. Methods There were 250 MI patients and 250 control subjects genotyped for 5 SNPs of the human CYP4F2 gene (rs3093105, rs3093135, rs1558139, rs2108622, rs3093200). Data were assessed on 3 separate groups: the total subjects, men and women. Results For men, G allele was significantly higher in the MI patients than in the control subjects and the overall distribution of the haplotypes was significantly different between the MI patients and the control subjects (P=0.002). Also in men, the frequency of T-C-G haplotype was significantly higher for MI patients than for control subjects (P=0.002), and the frequency of T-C-A haplotype was significantly lower for M1 patients than for control subjects (P=0.003). Conclusion Data from the present results indicated that MI was associated with G allele of rs2108622 in men, suggesting that T-C-G haplotype might serve as genetic marker for MI in men.
